Pasmo pass good for travel from Tokyo to Kamakura via Odakyu, Enoden, JR Kamakura and Tokyo subway?

Can I use a Pasmo card to travel with this itinerary?

From: Shinjuku Station on Odawara Line Express to Odawara to Sagami-Ono. Transfer to Enoshima Line Express to Fujisawa to Fujisawa.
Then take the Enoden train to Kamakura.

Return via JR Train from Kamakura station via JR Yokosuka Line to Shimbashi then transfer to Toei Subway Asakusa Line to Tobu Asakusa.

If so, do I just load the full amount of money needed and go to the trains and scan the card. If we would like to upgrade to a green seat on the return trip from Kamakura to Shimbashi, do I just add more money on the card?

You should be able to use the pasmo card for all those trips according to this website:

Also you can use the pasmo card on the enoden: http://www.city.kamakura.kanagawa.jp/kamakura-kankou/en/transportation/

For green seats you will probably have to pay more. I'd advise checking at the train station once in Japan.
