Depends on how you define "best". You need to catch a bullet train from Shinagawa Stn to go to Shin-Osaka Stn. To get to Shinagawa, you can take a Keikyu train, which is not covered by the pass - but it'll get you there slightly faster, and the fare is not much - just catch the train for the 25 minute ride (¥410). Alternatively, you can do it solely using the pass, though it'll take a bit longer. You take the monorail from the airport to Hamamatsucho, and then take a JR Yamanote Line train to Shinagawa. It's your choice.
